Police are investigating the double fatality, which occurred on Thursday at about 9.40am.
A police spokesman said a man and woman from one vehicle died at the scene.
Their ages are not yet known.
Officers from Far South Coast Local Area Command have established a crime scene and are investigating the circumstances surrounding the crash.
A report will be prepared for the Coroner.
The Snowy Hydro SouthCare rescue helicopter was sent to the scene.
The highway is closed, with all traffic diverted via Broulee Road and North Head Road.
Paramedics, police, SES and Rural Fire Service crews are on the scene about five kilometres north of Moruya.
The accident occurred at about 9.40am.
A NSW ambulance service spokesman said it was understood three vehicles were involved.
Police are diverting north-bound traffic via North Head Road, just north of the Moruya River bridge.
Southbound traffic is being diverted via Broulee Road.
The accident is understood to be near Waldrons Swamp.
It follows an accident involving a car and motorbike in the vicinity last month.
It is the second Princes Highway crash in two days, with two vehicles lodged on the median strip near the Batemans Bay Industrial area on Wednesday morning.
